SUBCELLULAR LOCALIZATION AND STUDY OF THE EXPRESSION OF THE METALOPROTEASE AAA FTSH-P1 IN Arabidopsis thaliana

AAA Proteins (ATPases Associated to different cellular Activities) are involved in important cellular processes. AAA family is classified in at least 6 subfamilies, being the FtsH subfamily belonging to a group of metalo-proteases. The eukaryotic FtsH proteases falls into five phylogenetic-related groups composed exclusively by either mitochondrial- (FtsH-m1 and FtsH-m2) or plastid-localized proteins (FtsH-p1, FtsH-p2 and FtsH-p3) that are involved in crucial cellular processes as oxidative phosphorylation, photosynthesis, and biogenesis of mitochondria and plastids. However, gene regulation of the encoded proteins as well as its function in plants, are still poorly understood. Therefore, the Arabidopsis FtsH-p1 protease gene was characterized and its promoter region was searched for cis-acting regulatory elements. The subcellular localization of the FtsH-p1 in vivo was performed by fusion of its targeting sequence to the Green Fluorescent Protein (GFP). This study validated its plastidial subcellular localization. Furthermore, in silico analysis of the expression of FtsH-p1 indicated both tissue-specific expression and salicylic acid induction of expression. We compared those findings with the promoter region analysis, revealing some insights into the effects of regulatory elements on expression of FtsH-p1. Which led us to perform an in vivo FtsH-p1 expression study. The promoter region of this gene was cloned into pCAMBIA-1301Z binary vector in order to regulate uidA (β-glucuronidase, GUS) gene expression. The constructions were introduced into Agrobacterium tumefaciens and further used to transform Arabidopsis plants.
