COVALENT IMMOBILIZATION OF TRYPSIN ONTO POLYANILINE | ||

INTRODUÇÃO:
Enzyme immobilization is a group of techniques that promote low cost and high stability of enzyme preparations. This is a great advantage for industries applications in large scale. The main theme of this essay is trypsin immobilization having polyaniline as support. |

METODOLOGIA:
The system was tested having as goal to optimize the immobilization conditions and reaction of the system with the casein substrate. As a way to optimize the immobilization parameters, the pH was tested, so as the time and concentration of the enzyme solution. However, in the reaction parameters were tested the pH, time and temperature. |

RESULTADOS:
After the tests, it was verified that the immobilized enzyme displays activity optimun related in a 7.6 immobilization pH, 60 min of immobilization time and concentration of 0.5 mg/mL. Reaction pH of 8.6 was obtained, in a 60 min period at 45º C. |

CONCLUSÕES:
This way was concluded that the immobilized enzyme displays a minimun diference if compared to the free enzyme in what is related to the conditions optimums of activity. Besides that the system was able to contain a large part of the free enzyme in a range of 97%. |
