EFFECT OF DOXORUBICIN AND VINBLASTINE IN EARLY EMBRYONIC DEVELOPMENT OF THE SEA URCHIN Echinometra lucunter

INTRODUÇÃO:

ABC superfamily of proteins have been widely studied since it was established its role in multidrug resistance phenotype (MDR). These proteins are involved in the transport of several xenobiotics and endogenous substances. ABC proteins act as efflux pumps in an energy-dependent mechanism that results in a reduction of the intracellular concentration of their substrates. The studies on the biological role of these proteins in gametes and embryonic cells are still incipient. Souza and colleagues (2009) demonstrated that the sea urchin Echinometra lucunter eggs accumulated more calcein-AM, a fluorescent substrate of ABCB1 and ABCC1 proteins, than embryonic cells, suggesting a higher ABC protein activity in embryos. ABC proteins modulators and substrates have helped in the understanding of the biological role of this family of proteins in a wide range of cell types. The objective of the present work was to investigate the effect of two chemotherapeutic agents (doxorubicin and vinblastine - also ABC substrates) in the embryonic development of E. lucunter.
METODOLOGIA:
Sea urchins were collected at Cabo Branco beach (João Pessoa/PB). Gametes were obtained by intracoelomic injection of KCl (0.5 M). Fertilization was done by adding one part of spermatozoa suspension in 99 parts of eggs suspension. Two kinds of protocols were applied: a) embryos were treated with different concentrations of doxorubicin or vinblastine 5 minutes after fertilization; b) eggs were treated with chemotherapeutic drugs 15 minutes before fertilization. At different time intervals, samples were fixed and development stage analysed under optical microscopy.
RESULTADOS:
Both chemotherapeutic drugs inhibited E. lucunter embryonic development independently of protocol used.  However, vinblastine was more effective than doxorubicin. Additionally, EC50 values (doxorubicin and vinblastine) were lesser when drugs were added to eggs before fertilization.
CONCLUSÃO:

These results may be attributed to the lower functional activity of ABC proteins in gametes than in embryonic cells of E. lucunter.
